A CRISPR-Cas9–engineered cell line carrying a mutation in the R2-box region of the R2 gene (within the ERE-homologous 5′UTR) exhibits markedly reduced R2 transcript and protein levels. These cells display slower growth and severely impaired HBV infection and replication, making them a valuable model for studying R2 regulation, HBV infection, and pathogenesis.
